We find ourselves in pandemic times. The global population is under siege by an infectious virus new to humankind. It’s called Severe Acute Respiratory Syndrome Coronavirus 2, or SARS-CoV-2. It’s the causative agent of the pandemic disease designated COVID-19. This viral adversary knows no politics. It recognizes no national boundaries. It is unconcerned with anyone’s identity. All 7.8 billion of us are the same to the virus: we are all hosts suitable to commandeer to make copies of itself.

The Biological Technologies Office is holding a Proposers Day meeting covering the new Detect It with Gene Editing Technologies (DIGET) program. DIGET aims to incorporate gene editors into detectors for distributed health biosurveillance and rapid, point-of-need diagnostics for endemic, emerging, and engineered pathogenic threats. The DIGET vision incorporates two devices: a handheld, disposable point-of-need device that screens samples for at least 10 pathogens or host biomarkers at once, combined with a massively multiplexed detection platform capable of screening clinical and environmental samples for more than 1,000 targets simultaneously.

DARPA’s Biological Technologies Office is hosting a Proposers Day meeting to provide information to potential applicants on the structure and objectives of the new PReemptive Expression of Protective Alleles and Response Elements (PREPARE) program. PREPARE will explore ways to better protect against biological, chemical, or radiological threats by temporarily and reversibly tuning gene expression to bolster the body’s defenses against—or directly neutralize—a given threat. It will focus on four key health challenges as proofs of concept: influenza viral infection, opioid overdose, organophosphate poisoning, and exposure to gamma radiation.
Pathogens with pandemic potential, toxic chemicals, and radioactive materials all endanger public health and pose a threat to national security. Despite investment in the development of medical countermeasures (MCMs) to address these threats, many existing MCMs suffer from limited applicability, insufficient efficacy, requirements for repeat dosing, lengthy and complex manufacturing processes, and logistically burdensome storage requirements. In many cases, unique threats require unique responses, setting up a “one threat, one MCM” paradigm.

Dr. Blake Bextine joined DARPA as a Program Manager in March 2016. He is interested in novel approaches to addressing issues facing agricultural biotechnology and biosecurity, including nucleic acid-based anti-pathogen/anti-pest strategies, insect and plant transformation strategies for rapid trait selection, and innovative approaches for altering insect behaviors.
