The Engineered Bio-Molecular Nano-Devices/Systems (MOLDICE) program has developed and demonstrated novel hybrid (biotic-abiotic) nanoscale interface technologies that enable direct, real-time conversion of bio-molecular signals into electrical signals. The transduction platform consists of a biological ion channel protein incorporated into a rugged artificial lipid bilayer. Low-noise, high-gain electronic amplifiers and custom software allow the detection and analyses of ionic current flowing through the ion channel. The protein channel itself has been modified via site-directed mutagenesis so that it specifically interacts with organophosphate molecules. When these molecules are present, current through the channel is transiently interrupted. Characteristic temporal patterns in the current blockages permit the blocking molecule, as well as its concentration, to be identified. The entire platform is portable and operates in an office environment without the need for electrical or vibration isolation.
